Hallo,
waarom is het belangrijk dat er bij een northern blot methyleen blauw gebruikt wordt voor de kleuring van het membraan? Ik had op internet gelezen dat dat is omdat dit niet intercaleert met het RNA zoals ethidium bromide dit wel doet. Maar wat is daar dan het gevolg van?

als dat intercaleert met het dna of rna dan gaat dat niet meer zo goed functioneren denk ik?
Maar wat voor kleuring doet methyleenblauw precies? want ik vind wel dat dat veel gebruikt wordt in de microbiologie, maar ik weet niet wat dat precies doet en waarom het dan zo nuttig is om dat te gebruiken bij een northern blot

Precies! Ethidium bromide is een mutagen kan aanleiding geven tot mutaties. Van het moment dat je ethidium bromide aan je staal toevoegd kan je niet meer voor 100% zeker zijn dat je RNA nog functioneel is.

Methylene Blue Stain is an aqueous solution of methylene blue (0.02%) designed for quantitative or qualitative examination of RNA and DNA immobilized on hybridization membranes in northern, Southern and alkaline blotting. It can be used to verify the amount of RNA on hybridization membranes in northern blotting. Unlike the carcinogenic agent, ethidium bromide, Methylene Blue Stain does not intercalate in nucleic acid chains and accordingly, does not interfere with nucleic acid retention on hybridization membranes nor with the hybridization process. Methylene Blue Stain was tested and found applicable for use with both plastic and nitrocellulose membranes. However, the nitrocellulose membranes exhibited higher background staining.
Methylene Blue Stain detects > 20 ng RNA or DNA/band.
