Beste,
wij zouden graag een bestaande gradient-methode optimaliseren. Een eerste stap hierbij is het bepalen van de Van Deemter-curve.
Echter, wij zijn niet zeker als we de bestaande gradient mogen blijven gebruiken of moeten terugschakelen naar een isocratische methode voor de bepaling van het minimum ?
Is de gevonden optimale flow bij een isocratische methode ook de optimale flow bij de gradient methode ?

Nee! Zie hieronder waarom (uit Perkin-Elmer handleiding overgenomen):
"The optimal flow rate in isocratic mode for a 2.1 mm I.D. column packed with 1.7 μm particles is around 500 μL/min. Due to the low mass transfer resistance generated by small particles (because of the reduced diffusion path)! When dealing with larger molecules, the mobile phase flow rate should be reduced to 300 μL/min due to a reduction of diffusion coefficients."
"The rules in gradient mode are different, where the highest flow
rate always provides the best peak capacity because the peak capacity
is dependent on the columns dead time and, to a lesser extent, on the columns
efficiency. Therefore, the flow rate for gradient HPLC experiments should be elevated, but at the maximum equal to 80-90% of the maximum pressure withstood by the instrument."
