IHC

Ik moet een protocol samenstellen voor een IHC aan de hand van een artikel, maar ik heb wat moeite met het interpreteren van het artikel. Kan iemand zeggen wat er nog ontbreekt en waar ik eventueel fout zit?

Protocol IHC volgens het artikel:
1. CIT (tijd tussen de weefselafname en fixatie)

2. Fixatie: fixatief / fixatietijd
a. Fixeer het longweefsel met 4% paraformaldehyde
b. Fixatietijd is onbekend
3. Dikte van de weefselcoupe
a. Versnijd het weefsel in coupes van 5µm
b. Fixeer het longweefsel met 4% paraformaldehyde
c. Permeabiliseer met 0.2% Triton in PBS.
d. Blokkeer met 10% goat serum in PBS

4. Keuze draagglas
a. Onbekend
5. Controles
a. Geen controles aanwezig
6. Keuze antigen retrieval: tijd / T / pH
a. Onbekend

a. Incubeer overnacht met de primaire antilichamen: N-cadherin, E-cadherin, α-SMA, MMP-9, vimentin en TGF-β bij 4°C.
b. Incubeer hierna met “HRP-conjugated goat anti-rabbit IgG secondary” antilichamen volgens de kit (ZSGB-BIO)

"Histology and immunohistochemical analysis:"

Lungs were inflated with 4% paraformaldehyde, embedded in par-affin and cut at a thickness of 5μm. For hematoxylin and eosin staining,sections werefixed in 100% methanol and stained according to astandard protocol. For immunohistochemical, sections werefixed with4% paraformaldehyde, permeabilized with 0.2% Triton in PBS, blockedwith 10% normal goat serum in PBS for 1 h. The sections were in-cubated with primary antibodies N-cadherin, E-cadherin,α-SMA(Abways, CY5295), MMP-9 (Santa Cruz Biotechnology, SC-13520),Vimentin (Proteintech, 10366-1-AP) and TGF-β(Cell SignalingTechnology, 3711), 4 °C, overnight followed by incubation with HRP-conjugated goat anti-rabbit IgG secondary antibody according to the kitFig. 4.HF decreased HDM-induced metaplasia of mice epithelial and collagen deposition in mice lungs. (A) Chemical staining for hematoxylin and eosin wasperformed in 5μm-thick lung sections from each group to examine inflammatory infiltration and mucus secretion. instructions (ZSGB-BIO). Peroxidase conjugates were subsequently vi-sualized by utilizing diaminobenzidine (DAB) solution. The sectionswere counterstained with haematoxylin and mounted on a coverslip.Tissues were imaged using Olympus microscope (Mantra, PerkinElmer)
